Elucidating the role of the gut-brain-axis in autism AutismResearch GutBrainAxis ASDPhenotypes Microbiome Metabolic DietaryPatterns Inflammation ASDInsights Neurodevelopmental NatureNeuro

By Pooja Toshniwal PahariaJun 28 2023Reviewed by Benedette Cuffari, M.Sc. In a recent study published in the journal Nature Neuroscience, researchers present a framework to utilize multi-omic data from well-defined groups and assess the contribution of gut-brain axis disruptions in the pathogenesis of autism spectrum disorder .

The gut microbiome plays a central role in regulating neuroimmune networks, modifying neural networks, and directly communicating with the brain. The metagenomic and metabolic contributions of the microbiome are crucial for understanding the functional framework of ASD. In addition, 16S rRNA sequencing differentials were calculated from a fecal matter transplantation study, including 18 pediatric ASD patients. In addition, a microbiome-diet co-occurrence analysis was performed.

Study findings Age and sex matching enhanced ASD data analysis. Differential ranking analysis revealed strong ASD-microbiome links. A functional architecture along the GBA correlated with the ASD phenotypic heterogeneity. This was characterized by ASD-associated lipid, amino acid, and carbohydrate profiles mainly encoded by Bifidobacterium, Prevotella, Bacteroides, and Desulfovibrio species and correlated with brain gene expression changes, pro-inflammatory cytokine profiles and restrictive diet patterns.

ASD phenotypes were also strongly associated with temporal gut microbial compositional alterations. In ASDs, interleukin-6 levels were upregulated, whereas Bifidobacterium, Prevotella, and Bacteroides counts were mainly associated with the cytokine differentials.
