By Neha MathurJul 13 2023Reviewed by Lily Ramsey, LLM In a recent article published in The Lancet Psychiatry, researchers analyzed the relationships between polygenic liability for bipolar disorder, schizophrenia, and major depressive disorder to stratify heterogeneous, multidimensional symptoms of bipolar disorder.

Another intriguing observation comes from family studies suggesting that there is independent transmission of the core symptom dimensions of bipolar disorder, MDD, and psychosis. Thus, despite their clinical concordance, they seem to be etiologically distinct. About the study In the present study, researchers extensively searched multiple databases, viz., MEDLINE, and PsychINFO, to name a few, and systematically identified all relevant literature published in English between January 1, 2006, and September 15, 2022, that used contemporary approaches for genomic stratification of mood disorders.

Related StoriesInclusion criteria for enrolment into this study were that individuals were 18 years or older at the time of the interview, were of European ancestry , lived in the United Kingdom , and registered with the BDRN. The multiple indicator multiple causes model allowed researchers to explore the patterns of complex associations between polygenic liability indexed by disease symptoms, their dimensionality, and polygenic risk scores .

Results In total, 4,198 individuals were eligible for inclusion in this study; however, only 4,148 individuals of EA living in the UK with a median age of 45 years at the SCAN interviews constituted the analysis sample set, of which 2,804 and 1,344 were female and male, respectively. Conclusions The study results challenged current diagnostic systems and highlighted that dimensional representations of bipolar disorder could accommodate problems related to poorly explained mixed psychiatric states with the same symptom presentation.

Immunotherapy has attracted attention as a method to prevent this condition. However, the conventional immunotherapy method targeting solid tumors, that is, intravenous injection, has limitations in homing in on the tumor and in vivo expansion and has not shown effective clinical results. Method To overcome these limitations, NK cells (Natural killer cells) were encapsulated in micro/macropore-forming hydrogels using 3D bioprinting to target solid tumors. Sodium alginate and gelatin were used to prepare micro-macroporous hydrogels. The gelatin contained in the alginate hydrogel was removed because of the thermal sensitivity of the gelatin, which can generate interconnected micropores where the gelatin was released. Therefore, macropores can be formed through bioprinting and micropores can be formed using thermally sensitive gelatin to make macroporous hydrogels. Results It was confirmed that intentionally formed micropores could help NK cells to aggregate easily, which enhances cell viability, lysis activity, and cytokine release. Macropores can be formed using 3D bioprinting, which enables NK cells to receive the essential elements. We also characterized the functionality of NK 92 and zEGFR-CAR-NK cells in the pore-forming hydrogel. The antitumor effects on leukemia and solid tumors were investigated using an in vitro model. Conclusion We demonstrated that the hydrogel encapsulating NK cells created an appropriate micro–macro environment for clinical applications of NK cell therapy for both leukemia and solid tumors via 3D bioprinting. 3D bioprinting makes macro-scale clinical applications possible, and the automatic process shows potential for development as an off-the-shelf immunotherapy product.
